Anti-CD3 mAb delays or prevents type 1 diabetes (T1D) by modulating the immune mediated destruction of beta cells. Our findings described the reshaping of islet-infiltrating T cells and beta cells that lead to operational, but tenuous tolerance to autoimmune diabetes following anti-CD3 mAb treatment. Overall design: Islets and paired pancreatic draining lymph nodes were isolated from anti-CD3 treated NOD mice in remission and processed for single-cell 5' RNA sequencing. Each scRNAseq dataset is multiplexed and contains islet cells or pancreatic draining lymph nodes of 3 mice, each individually labeled by hashtag barcode.
