First draft genome sequence of the rock bream in the family Oplegnathidae

The rock bream (<i>Oplegnathus fasciatus</i>) is one of the most economically valuable marine fish in East Asia. Various factors including temperature changes, pathogenic infections, and disorders related to hereditary and somatic mutations have led to huge revenue losses in production. In order to attempt to overcome these issues on the molecular scale, reference genome sequences are needed. In this study, we constructed the first draft genome for the family <i>Oplegnathidae (O. fasciatus) </i>to develop genetic bio-markers for the above-mentioned problems. The <i>O. fasciatus </i>genome size was estimated to be 780 Mb based on 235 Gb of fragmented sequences that were assembled into a 766-Mb draft genome. From the draft genome, 23,338 genes were predicted and the gene coverage was estimated by comparing the core eukaryotic genes. In total, 81.9% of core eukaryote genes were present in the total gene contents. Moreover, 97% of the genes were obtained the functional terms from similar fish genomes. To the best of our knowledge this is the first draft genome for the family <i>Oplegnathidae</i>.

条石鲷(Oplegnathus fasciatus)是东亚地区极具经济价值的海水鱼类之一。温度变化、病原感染以及遗传与体细胞突变相关的紊乱等诸多因素,已给该鱼种的养殖生产造成了巨额经济损失。为从分子层面攻克上述难题,亟需获取参考基因组序列。本研究构建了石鲷科(Oplegnathidae,O. fasciatus)的首个草稿基因组,以期为解决上述问题开发遗传生物标记物。基于235 Gb的片段化测序序列,我们预估条石鲷的基因组大小为780 Mb,并将其组装为766 Mb的草稿基因组。从该草稿基因组中,共预测得到23338个编码基因,并通过比对核心真核基因集评估了基因覆盖度:总计81.9%的核心真核基因可在该物种的全部基因集合中被检出。此外,97%的基因可通过比对其他相似鱼类基因组获得功能注释信息。据我们所知,本研究是石鲷科的首个基因组草稿。
