Impact of Vitamin-D3 on the gene expression profile of mouse colon

Vitamin-D3 (VitD3) exhibits pleiotropic effects in hosts physiology by acting on several organs. To mechanistically understand how VitD3 influences the colon physiology we interrogated the gene expression profile of mouse colon in response to high or low VitD3 bioavailability. VitD3 tissue bioavailability was modulated in mice with the use of diets supplemented with different concentrations of VitD3 (0, 2 or 10 IU/g) and the use of Gc knockout mouse strain in where VitD3 bioavailability is enhanced. RNA will be extracted from 5 biological replicates (5 individual mice) per condition (20 samples in total). Male mice (8 weeks old) were fed with the following diets 3 weeks :1)C57BL/6J/WT mice that were fed with standard VitD3 diet (2 IU/g) for 3 weeks (n=5), 2) C57BL/6J/WT mice that were fed with high VitD3 diet (10 IU/g) for 3 weeks (n=5), 3) Gc KO mice that were fed with standard VitD3 diet (2 IU/g) for 3 weeks (n=5), 4) Gc KO mice that were fed with VitD3 deficient diet (10 IU/g) for 3 weeks (n=5)

维生素D3(Vitamin-D3,VitD3)可通过作用于多个器官,对宿主生理产生多效性调控作用。为从机制层面解析VitD3对结肠生理的调控机制,本研究针对不同VitD3生物利用度条件下的小鼠结肠基因表达谱开展了分析。我们通过两种方式调控小鼠体内VitD3的组织生物利用度:其一为饲喂添加不同浓度VitD3(0、2或10 IU/g)的饲料;其二为使用VitD3生物利用度升高的Gc基因敲除(Gc knockout)小鼠品系。 本研究的每个实验条件均设置5个生物学重复(即5只独立小鼠),总计20个样本。实验选用8周龄雄性小鼠,分别进行为期3周的饲喂处理,具体分组如下: 1. C57BL/6J野生型(wild type, WT)小鼠,饲喂标准VitD3饲料(2 IU/g),共5只; 2. C57BL/6J野生型(WT)小鼠,饲喂高剂量VitD3饲料(10 IU/g),共5只; 3. Gc基因敲除小鼠,饲喂标准VitD3饲料(2 IU/g),共5只; 4. Gc基因敲除小鼠,饲喂VitD3缺乏饲料(10 IU/g),共5只。
