To quantify fitness across diverse conditions, we performed barcode amplicon sequencing on a pool of 495 DNA barcoded Saccharomyces cerevisiae isolates. This pool was grown competitively in 704 distinct conditions and harvested at fixed timepoints. Genomic DNA was extracted, and barcode regions were amplified in a two-step PCR protocol to prepare libraries for high-throughput sequencing.
