Trichoplusia ni cell lines are commonly used to enable recombinant protein expression via baculovirus infection, and have been used to generate materials currently approved for clinical use or in clinical trials. In order to develop systems biology and genome engineering tools to improve protein expression work in this host, we performed a genome assembly of Trichoplusia ni using the Tni-FNL cell line derived from T. ni BTI-Tn-5B1-4. We created 3 sequencing library types including PacBio long read library, 10x genomics linked reads library and Bionano Irys optical mapping library. We sequenced genomic DNA and assembled the Tni-FNL genome using a hybrid approach to build genome assemblies and scaffolds. This genome assembly has N50 scaffold size of 2.3Mbp and a total length of 359 Mbp.
