Lactic Acid Bacteria (LAB), which are widely used in functional foods, have attracted extensive research attention in recent years. Lactic acid bacterial strains that release bioactive antihypertensive peptides have been the focus of exploration. In this study, 67 LAB strains were isolated from traditional Daqu. Fifty strains with angiotensin-converting enzyme inhibitory (ACE-I) activity exceeding 16% were selected for 16S rDNA sequencing and safety evaluation. Notably, Enterococcus faecium CP640 and Lactobacillus rhamnosus CP2 exhibited high ACE-I inhibitory activity when fermented in reconstituted skim milk. These selected strains showed no hemolytic activity or antibiotic resistance, did not produce biogenic amines, and displayed high survival rates in the gastrointestinal tract. In addition, the fermented milk products of Enterococcus faecium CP640 and Lactobacillus rhamnosus CP2 significantly reduced blood pressure levels in Spontaneously Hypertensive Rats (SHR). No adverse effects were observed in normal Wistar-Kyoto (WKY) rats compared to the negative control group. Through physiological, serum and biochemical analyses, Enterococcus faecium CP640 and Lactobacillus rhamnosus CP2 were found to have potential beneficial effects including promoting weight gain in SHRs, relieving tachycardia, improving renal function indicators, and effectively regulating blood glucose and uric acid levels in SHRs. These two strains exhibit optimal antihypertensive properties and are suitable for development into functional dairy products.
