Human osteosarcoma xenografts were implanted adjacent to the lateral femur (at mid point, diaphysis) of NOD/SCID mice. In the DkkMO group, a Dickkopf-1 blocking morpholino was injected intraperitoneally every 2 days (12.5 mg per kg). Controls received vehicle. After 14 days, mice were humanely euthanized. Tumors and associated stroma were recovered and cells were processed for single-cell sequencing.
