The brain is an important controller of glucose homeostasis and mediates counterregulatory responses to hypoglycemia. We used single-nuclei transcriptomics to study the mechanisms of defective hypoglycaemia counter-regulation in brain in a model of diabetic mice exposed to acute or recurrent hypoglycemia. Overall design: Single nuclei were isolated from hypothalamus and cortex tissues of diabetic mice subjected to recurrent (RH) or acute hypoglycemia (AH) and used to generate transcriptome profiles.
