A two-phase liquid differentiation of peripheral blood mononuclear cells (PBMC) can be used to study human erythroid cells. To determine the epigenetic landscape of erythroid cells derived from PBMC we have performs ChIP-seq for histone marks associated with promoters (H3K4me3), enhancers (H3K4me1) and polycomb repression (H3K27me3) as well as for chromatin bound pol2. In addition we characterized the binding of the boundary element CTCF and key erythroid transcription factors NF-E2 and SCL/TAL.
采用两步液体分化培养法对外周血单个核细胞(peripheral blood mononuclear cells, PBMC)进行处理,可用于研究人类红系细胞。为解析源自PBMC的红系细胞的表观遗传图谱,我们针对与启动子相关的组蛋白标记H3K4me3、与增强子相关的组蛋白标记H3K4me1、与多梳蛋白抑制相关的组蛋白标记H3K27me3,以及结合于染色质的RNA聚合酶II(Pol II)开展了染色质免疫共沉淀测序(ChIP-seq)。此外,我们还对边界元件CTCF以及关键红系转录因子NF-E2、SCL/TAL的结合特征进行了鉴定。
