MicroRNAs (miRNA) play key roles in diverse biological processes including heart development. In our study, we evaluated the effects of DHW oil on miRNA expression in mahi-mahi (Coryphaena hippurus) embryos exposed to WAF from weathered slick oil and non-weathered source oil. miRNA and mRNA was compared using downstream bioinformatics with subsequent target organ predictions based on their interactions. Using two methods miRNA sequences were annotated using the Fugu rubripes genome or using BLAST. Our data showed that exposure of embryos to slick oil resulted in more differentially expressed (DE) miRNAs than source oil at all developmental stages 24hpf, 48hpf and 96hpf. There was also an increase in the number of DE miRNA as development progressed, 96h having the highest number of DE miRNAs. Gene ontology (GO) analysis on the target mRNAs was consistent with pathways analysis of miRNAs, predicting disruption of cardiovascular system development after oil exposure and showed specific miRNA–mRNA interactions that may contribute to these effects. This work is the first study linking miRNAs and mRNAs in fish responsive to DWH oil exposure, providing a new opportunity for better understanding of the molecular mechanism of DWH oil toxicity.

微小核糖核酸(miRNA)在包括心脏发育在内的多种生物学过程中发挥关键作用。本研究评估了DWH油对暴露于风化浮油水相馏分(WAF)与未风化原油水相馏分的棘鳍鲯鳅(Coryphaena hippurus)胚胎中miRNA表达的影响。随后通过下游生物信息学方法对miRNA与信使核糖核酸(mRNA)进行比较,并基于二者的相互作用开展靶器官预测。本研究通过两种方法对miRNA序列进行注释:一是利用红鳍东方鲀(Fugu rubripes)基因组,二是采用BLAST序列比对工具。数据显示,在24hpf、48hpf及96hpf所有发育阶段,胚胎暴露于浮油后的差异表达(DE)miRNA数量均多于暴露于原油的胚胎;且随着发育进程推进,DE miRNA的数量逐渐升高,96hpf阶段的DE miRNA数量达到峰值。对靶mRNA进行的基因本体论(GO)分析与miRNA通路分析结果一致,均预测油暴露会干扰心血管系统发育,且揭示了可能介导此类效应的特异性miRNA-mRNA相互作用。本研究是首个探讨鱼类中miRNA与mRNA响应DWH油暴露关联的研究,为深入理解DWH油毒性的分子机制提供了全新的研究视角。
