In April 2024, at the morel mushroom (M. sextelata) cultivation park located in Shuangcheng District, Harbin City, Heilongjiang Province, China , certain greenhouses exhibited either dead or absent mushroom growth (Figure 1). To investigate the causes of these issues, a five-point sampling method was employed to collect surface soil (1-5 cm) from within the same greenhouse, which were then combined into a single sample. The soil samples from each location were sieved through a 2 mm sieve, with roots and plant debris manually removed. Subsequently, the sample was divided into two parts: one was air-dried for soil physicochemical property determination, and the other was rapidly placed in liquid nitrogen, transported to the laboratory, and stored in a -80 Celsius freezer for subsequent DNA extraction. A total of nine soil samples were collected, comprising successfully fruiting samples (CK, 3 samples), dead mushroom samples (DM, 3 samples), and unfruitful samples (UM, 3 samples).
