Wild-type KRAS activation drives evasion of interferon-mediated immunity and resistance to immunotherapy in hepatocellular carcinoma

Based on scRNA-seq analysis, we found that KRAS inhibited the expression of intrinsic interferon-mediated MHC-I and impaired extrinsic interferon-induced dendritic cell recruitment, resulting in a reduction of CD8+ T cell activity due to the suppression of CXCL9 through the EGFR/MEK/ERK signalling pathway. The tumor immune microenvironment of the c-MYC-lucOE/Tp53KO/EV and c-MYC-lucOE/Tp53KO/Kras mice were compared using scRNA-seq.
