HDBR project: 200636. Skin is the largest organ in the human body and is our first line of defense against microbes and external insults, yet the series of molecular events and cell types that give rise to a fully functional human skin and its appendages remain unclear. This has been partly due to techniques with limited resolution for characterization of skin development. As part of Human Cell Atlas initiative, in this study initially we will use single cell RNA + ATAC sequencing on fetal skin tissue dissociated into single cell suspensions. Subsequently, we will map the location of the identified cell types using spatial methods such as spatial transcriptomics, immunofluorescence and spatial CITE-seq.This will inform us about the developing human skin across multiple developmental stages which will also inform generation of in vitro skin organoid models.
