Antibiotic resistance is one of the greatest threats to human health nowadays. The success of antimicrobial therapy is influenced by characteristics of the infecting pathogen, which allow them to evade the effects of antibiotics, as well as patients’ characteristics such as renal function and dialysis status. It is important to take these factors into account when choosing optimised dosing regimens of antibiotics to ensure infection resolution. This thesis includes experimental model, novel quantitative and systems pharmacology modelling, population pharmacokinetic modelling and simulations to provide insights into optimising antibiotic therapies to maximise bacterial killing and minimise the emergence of resistance.
