The aim of this study was to generate a new, high-quality <em>Tribolium castaneum</em> genome assembly (TcasONT) by combining Oxford Nanopore long-read sequencing, and reference-guided assembly approach. The new TcasONT assembly proved to be enhanced by nearly 53 Mb compared to the last version of the <em>T. castaneum </em>genome, Tcas5.2. The TcasONT assembly exhibits higher gene completeness and a 20-fold enrichment in the repetitive genome part. Indeed, the enlargement of the TcasONT assembly was up to 50 Mb when all classes of repetitive sequences were considered. Therefore, TcasONT assembly not only contributes to the identification of potential novel genes but also provides an excellent platform for the analysis of different repetitive genome fractions.

本研究旨在结合牛津纳米孔(Oxford Nanopore)长读长测序技术与参考基因组引导组装策略,构建一套全新的高质量赤拟谷盗(Tribolium castaneum)基因组组装版本TcasONT。相较于赤拟谷盗上一版基因组组装Tcas5.2,新版TcasONT组装的基因组规模提升了近53 Mb。TcasONT组装的基因完整性更佳,且重复基因组区域的富集度达到20倍。实际上,若统计所有类别的重复序列,TcasONT组装的基因组增幅可达50 Mb。因此,TcasONT基因组组装不仅可助力潜在新基因的鉴定,更为不同重复基因组组分的分析提供了优质的研究平台。
