This exon array analysis was performed on mice carrying splice specific mutations at the end of exon9 of the Wilms tumor supressor gene that interfere with the production of WT1(+KTS) isoforms (Hammes et al., 2001; Cell 106:309) 4 wildtype and 4 wt1(+KTS) mutant E18.5 mouse embryos were transcardially perfused with magnetic beads, glomeruli isolated and RNA extracted and analyzed for differential expression on Affymetrix Exon Arrays
本研究针对携带威尔姆斯肿瘤抑制基因(Wilms Tumor Suppressor Gene, WT1)外显子9末端剪接特异性突变的小鼠开展外显子芯片(exon array)分析,该突变会干扰WT1(+KTS)亚型的产生(Hammes等人,2001;《Cell》106卷:309页)。我们对4只野生型小鼠和4只携带wt1(+KTS)突变的E18.5小鼠胚胎以磁珠溶液经心脏灌流,随后分离肾小球,提取RNA,并通过Affymetrix外显子芯片进行差异表达分析。
