Proteomic Characterization of Circulating Tumor Cells (CTCs) by Image Mass Cytometry (IMC)

CTCs from bone marrow aspirate sample of a metastatic prostate cancer were analyzed using IMC. There are total 40 region of interests (ROIs) included for proteomic charaterization as txt files. Based on those files, cell segmentation and ion count per cell could be generated by IMC segmentation pipeline from the Bodenmiller Lab, using the CellProfiler and Ilastik. Cell images with segmented masks could be further visualized by HistoCAT. MCD file is for a quick overview on MCD Viewer.

本研究采用成像质谱流式细胞术（Imaging Mass Cytometry, IMC）分析了转移性前列腺癌骨髓抽吸样本中的循环肿瘤细胞（Circulating Tumor Cells, CTCs）。该数据集共包含40个用于蛋白质组学表征的感兴趣区域（ROIs），以文本文件（txt files）格式存储。基于上述文本文件，可通过博登米勒实验室开发的IMC分割流程，结合CellProfiler与Ilastik软件完成细胞分割并计算单细胞离子计数。带有分割掩码的细胞图像可通过HistoCAT进行可视化。MCD文件可通过MCD查看器（MCD Viewer）实现快速概览。
