The ultimate step of tRNA wobble uridine modification 5-methoxycarbonylmethyluridine (mcm5U) and 5-methoxycarbonylmethyl-2-thiouridine (mcm5s2U) is generated by ABH8. Four tRNAs have been presented to bear the ABH8-catalyzed modification. In this study, we provide a global view on the binding repertoire of human ABH8 by HITS-CLIP and RIP-seq of ABH8-bound RNA. We demonstrate tRNA-Lys-UUU as a target that has not been previously identified in humans.
