The purpose of this experiment was to analyze the expression of NK cell ligands after their encoding genes were individually knocked out with CRISPR. We analyzed the level of ligand expression in both HIV-infected (p24+) and mock-infected CD4+ T cells. Conclusion: We noted an expected reduction in the expression level of each ligand indicated by the mean fluorescence intensity of flow cytometry staining in targeted knockouts compared to unedited CD4+ T cells both in total CD4+ T cells as well as p24+ HIV-infected cells. This indicates that CRISPR editing targeted each gene-of-interest as expected.
