<p>Surface sediment samples were collected for potential nitrification rates, using sediment slurries with filtered site water. Ammonia oxidizer gene abundances (AOA and AOB <em>amoA</em>) were quantified using qPCR, and clone libraries for each gene were sequenced using Sanger sequencing.</p>

本研究采用经采样位点过滤后的水体制备沉积物悬液,采集表层沉积物样本以测定潜在硝化速率。采用实时定量聚合酶链反应(quantitative Polymerase Chain Reaction,qPCR)对氨氧化古菌(AOA,Ammonia-oxidizing Archaea)与氨氧化细菌(AOB,Ammonia-oxidizing Bacteria)的amoA基因丰度进行定量,并针对两类基因分别构建克隆文库,采用桑格测序(Sanger sequencing)完成测序。
