The Essentukskoye mineral water deposit (EMWD) is a part of the Mineralovodskiy artesian basin located in the Ciscaucasia (Russia). We have sampled this mineral water directly via the supply wellbore and set up enrichment cultures with fresh water samples and a number of mineral electron acceptors and organic and inorganic electron donors in order to assess the diversity of culturable prokaryotes in the aquifer. In the primary enrichment with an Fe(III) mineral ferrihydrite as the electron acceptor and acetate as the electron donor, 65% of 16S rRNA gene library was represented by a single phylotype falling into the Deferribacterales order as the novel family-level branch. From this enrichment, a pure culture of an anaerobic moderately thermophilic, neutrophilic, halotolerant motile vibrio, designated as strain Es71-Z0220, was isolated. The organism obligately depends on the presence of carbonate in the culture medium, it reduces synthesized ferrihydrite or fumarate as the electron acceptors using acetate, lactate, succinate, or pyruvate as the electron donors. It is unable to ferment peptides, carbohydrates or amino acids. Phylogenetic analyses based on 16S rRNA gene sequence and sequences of 120 bacterial single copy conserved marker proteins placed the strain Es71-Z0220 on a separate family-level branch within the order Deferribacterales. On the basis of its physiological characteristics and phylogenetic position the novel isolate is considered to represent a novel family, genus and species for which the following name Deferrivibrio essentukiensis gen. nov., sp. nov. strain Es71-Z0220 is proposed.
