To dissect the genomic function of Esr1 in neonatal cortical neurons, we performed estradiol benzoate or vehicle treatment on P1 Esr1-Cre;LSL-Sun1-GFP animals and harvested the neocortex to perform single-nucleus assay for transposase-accessible chromatin using sequencing (snATAC-seq) with the GFP+ cells. Esr1-Cre;LSL-Sun1-GFP pups at P1, 6hr post treatment of either vehicle or estradiol-benzoate injection, were used for this experiment. After tissue dissection and nuclei isolation, GFP+ and 7-AAD+ nuclei in each condition were collected into cold PBS with 0.1% BSA using Fluorescence-activated cell sorting (FACS). Collected nuclei were enriched with ConA beads and proceed to 10X single nuclei ATAC-seq (V2)
