Analysis of gonadal fat from male control and adipocyte-specific LAMA4 knockout mice

To determine the transcriptomic effects of LAMA4 loss on adipose tissue structure and gene expression in two dietary paradigms, we assessed gonadal white adipose tissue of male control and adipocyte-specific LAMA4 knockout mice using a Quant-Seq strategy. Epididymal fat pads were dissected from a) 6 male wild-type and 6 male adipocyte-specific LAMA4 knockout mice following 20 weeks of regular diet feeding, and b) 6 male wild-type and 6 male adipocyte-specific LAMA4 knockout mice following 20 weeks of 45% fat diet feeding, followed by isolation of total RNA and messenger RNA quantification via Quant-Seq.

为探究LAMA4缺失在两种饮食模式下对脂肪组织结构与基因表达的转录组学影响,我们采用Quant-Seq(Quant-Seq)策略,对雄性对照小鼠及脂肪细胞特异性LAMA4敲除小鼠的性腺白色脂肪组织进行分析。具体实验分组及取材如下:a) 对经20周正常饮食喂养的6只雄性野生型小鼠与6只雄性脂肪细胞特异性LAMA4敲除小鼠,剥离其附睾脂肪垫;b) 对经20周45%脂肪含量饮食喂养的6只雄性野生型小鼠与6只雄性脂肪细胞特异性LAMA4敲除小鼠,剥离其附睾脂肪垫。完成取材后,我们分离总RNA,并通过Quant-Seq技术对信使RNA进行定量检测。
