The goal of this study was to determine the genes regulated by ID4 in mammary epithelial subpopulations. Basal, luminal progenitor, and mature luminal mammary epithelial subpopulations were FACS-sorted from adult mice that were either WT or ID4 KO in quadruplicate. RNA-seq and differential gene expression analysis was performed. Overall design: Mammary glands were pooled from 4-8 WT or ID4 KO female adult (10-12 week) mice at estrus and digested into a single cell suspension. Live lineage negative CD24+CD29hiCD61+ basal, CD24+CD29loCD61+ luminal progenitor and CD24+CD29loCD61- mature luminal cells were isolated by FACS and RNA was extracted for RNA-seq. 4 biological replicates were performed.
