Gene expression in eosinophils isolated by FACS from inguinal subcutaneous adipose tissue of WT and mice lacking the transcriptional repressor Kruppel-like factor 3 (KLF3). We aimed to investigate deregulation of pathways and genes in the absence of KLF3 in eosinophils that reside in the subcutaneous adipose tissue, in order to study the role of KLF3 and its target genes in adipose eosinophil function and activity. Inguinal subcutaneous adipose tissue was dissected from 12-wk age- and litter-matched WT and KLF3 KO mice (n=3 per genotype). Adipose was digested using type II collagenase to yield the stromal vascular fraction (SVF), which was stained with fluorescently-conjugated antibodies to isolate eosinpohils (Live CD45+ CD11b+ F4/80+ SiglecF+ SSC hi). RNA was extracted from isolated cells using a Qiagen RNeasy Micro Kit then subjected to quality control using an Agilent 2100 Bioanalyzer, following preparation with an Agilent RNA 6000 Pico Kit. An Affymetrix 3’ IVT Pico Kit was used prior to microarrays which were performed on an Affymetrix GeneChip HT MG-430PM Array Plate.
