A. Protein lysates were harvested from fibroblasts infected with TB40/E wild type (WT), UL88-deficient, or UL88Rev HCMV at MOI of 3 at 96hpi. Blots were probed for viral proteins UL88, pp28, and tubulin as a loading control. Protein lysates from fibroblasts, either mock-infected or infected with wild type (WT) or UL88-STOP TB40/E strain of HCMV at a MOI of 0.05, were harvested at (B) 15 dpi or (C) 12dpi. The lysates were analysed by western blot (WB) and probed for MyD88, JAK1, NF-kB, IRF3, IRF1, p-STAT1, STAT1, EAA1, pp28, and tubulin. B-C. Graphs show densitometry analysis from 3 independent experiments for each set at 12 dpi. D. Protein lysates were harvested from fibroblasts infected with either UL88Revertant or UL88GalK virus at a low MOI of 0.05 and harvested at 12 dpi. Lysates were analysed by western blot and probed for MyD88 and tubulin (loading control). E. RNA was harvested at 12 dpi from fibroblasts infected with WT or UL88-STOP virus at low MOI of 0.05. A. 以感染复数(Multiplicity of Infection, MOI)为3,于感染后96小时(hours post infection, hpi)收集感染TB40/E野生型(WT)、UL88基因缺陷型(UL88-deficient)或UL88回复突变株(UL88Rev)人巨细胞病毒(Human Cytomegalovirus, HCMV)的成纤维细胞蛋白裂解液。以病毒蛋白UL88、pp28及作为内参的微管蛋白(tubulin)为靶标进行免疫印迹杂交检测。 从模拟感染(mock-infected),或感染MOI为0.05的TB40/E野生型(WT)或UL88基因终止突变株(UL88-STOP)HCMV的成纤维细胞中收集蛋白裂解液,收集时间分别为感染后15天(days post infection, dpi,对应B组)及12 dpi(对应C组)。采用蛋白质免疫印迹(Western Blot, WB)分析上述裂解液,靶标蛋白包括MyD88、JAK1、NF-κB、IRF3、IRF1、p-STAT1、STAT1、EEA1、pp28及微管蛋白(tubulin)。 B-C. 两组样本在12 dpi时的光密度分析结果均来自3次独立重复实验。 D. 以MOI为0.05的低感染复数感染UL88回复突变株或UL88GalK病毒的成纤维细胞,于12 dpi时收集蛋白裂解液。采用WB分析裂解液,以MyD88及微管蛋白(tubulin,内参)为靶标进行检测。 E. 以MOI为0.05的低感染复数感染野生型(WT)或UL88基因终止突变株(UL88-STOP)病毒的成纤维细胞,于12 dpi时收集总RNA。采用逆转录实时定量聚合酶链反应(Reverse Transcription Quantitative Polymerase Chain Reaction, RT-qPCR)分析MyD88 mRNA水平,以甘油醛-3-磷酸脱氢酶(Glyceraldehyde-3-Phosphate Dehydrogenase, GAPDH)作为内参进行标准化。结果呈现3次独立重复实验的倍数变化值,以平均值±标准差(mean ± SD)表示,*p<0.05为差异具有统计学意义。
