3' scRNA of R26-SETBP1-VAV fl/fl and R26-SETBP1-VAV-G870S mice

To investigate in vivo the role of SETBP1 somatic mutations in the onset and progression of TN-MF, we obtained a conditional mouse model to mimic the accumulation of mutated SETBP1 in the BM and in PB cells. A floxed human SETBP1G870S expression cassette, followed by an IRES-GFP sequence, was inserted in the safe-harbour Rosa26 locus (R26-SETBP1fl/fl; Sessa Lab). Crossing R26-SETBP1-G870S floxed mice with the hematopoietic specific Vav-iCre strain (Mead, Blood 2018) we obtained SETBP1G870S overexpression in the entire hematopoietic system. 3' 10x Single-cell sequencing was performed on CD34+ Lin- bone marrow cells on both mice.
