Unicycler assemblies

Unicycler assemblies (normal mode, version 0.4.7) using ONT and Illumina HiSeq reads. ONT reads were demultiplexed and basecalled using Guppy (version 4.2.3). Porechop (version 0.2.4) was considered to trim and remove ONT adapters with default parameters. Filtlong (version 0.2.0) was used to filter the ONT reads considering a minimum length of 1 kbp (--min_length 1000), a quality phred score of 20 (--mean_q_weight 20), retaining 90% of the total number of ONT reads ( --keep_percent 90) from a maximum 40x coverage (--target_bases).

本数据集采用ONT与Illumina HiSeq测序读段，通过Unicycler组装工具（版本0.4.7，正常模式）完成基因组组装。具体处理流程如下：ONT测序读段先使用Guppy软件（版本4.2.3）完成分样与碱基识别；随后采用Porechop软件（版本0.2.4），以默认参数修剪并移除ONT测序接头；最后使用Filtlong软件（版本0.2.0）过滤ONT测序读段，过滤参数设置为：最小长度1千碱基对（--min_length 1000）、平均质量Phred评分20（--mean_q_weight 20）、保留总读段数的90%（--keep_percent 90），且最大覆盖度为40倍（--target_bases）。
