To evaluate the effect of OCT1 in breast cancer cells, expression of OCT1 was knocked down in MCF-7 cells by small interfering RNA (siRNA). Microarray analysis was performed to analyze gene expression profiles of these cells. MCF-7 cells were treated with two specific siRNAs targeting OCT1 (siOCT1 #1 and siOCT1 #2) and a control siRNA targeting firefly luciferase (siControl). Total RNA was extracted from the cells for gene profiling 48 h after transfection of siRNA. RNA integrity number (RIN) was measured by Agilent 2100 Bioanalyzer (Agilent Technologies, Palo Alto, CA, USA). It was confirmed that RINs of all RNAs were above 8.6. RNAs extracted from biological triplicates treated with the same siRNA were combined, and subjected to the microarray analysis using Clariom S Array, Human (Affymetrix/Thermo Fisher Scientific, Walthan, MA, USA).
