For PacBio Isoseq RNA was extracted from whole Myzus persicae Clone O alate females using Trizol (Merck) and DNEasy (Qiagen). Libraires were prepared and sequenced on the PacBIO Sequel I instrument at the Earlham Institute, Norwick UK. For Illumina sequencing, RNA was isolated from dissected organs of Myzus persicae Clone O alate and winged females using Oligo-dT capture from lysate. Libraries prepared using Illumina Nextera and sequenced on an Illumina Hiseq 2500 at the Earlham Institute, Norwich UK.
