Burns are damage to the integrity of the normal skin caused by contact with a heat source and are destructive. The body's efforts to carry out the healing process have 3 phases, namely: inflammation, proliferation and maturation.Tissue damage from burns causes inflammation that causes pain.It requires a therapy that can be used in the process of epidermal re-epithelialization and reduce pain due to burns. In previous studies, it has been proven that the extract of the Bangun-bangun leaves (Coleus amboinicus) has effective as a reliever of pain and inflammation after acetic acid induction. In the Karo tribe, it is known as Daun Bangun-bangun (DBB). As a traditional medicine, it is often used directly on the injured skin. Aims: To analyze the effects of DBB extracts in topical gel to reduce pain and accelerate post-burn reepithelialization in rats. Methods: This is an experimental study design with a posttest-only control group design. The sampling method was randomized consisted of 54 rats divided into 9 groups, i.e:I.negative control;II.positive control (Bioplacenton®) and III.Treatment (36% DBB extracts gel). All rats were given second-degree burns except the standard group. The animal was treated 2x in daily. Each group was subdivided into three smaller groups (n = 6) according to the time the lesions were evaluated. Skin tissue was taken on days 3, 14, 21, and data were analyzed to compare the decrease in pain threshold between groups and to calculate the number of leukocyte migration in the healing process. If the data is normally distributed then continued with ANOVA and post hoc Bonferroni tests but, if the data is abnormally distributed, it continued with Kruskal Wallis test. Results: There were decreased of pain in EDBB and positive control (bioplacenton) groups, but in statistically no significantly different in reduced pain (p=0.139). The increasing of number of leucocyte migration compared of EDBB to other groups depend on the day of assessed, but in macroscopis we can see the best healing process in all groups is EDBB group although two other groups almost equally. Conclusion: Bangun-bangun leaves (Coleus amboinicus) extracts gel could reduce pain and accelerate post-burn reepithelialization in rats.
