We have developed a minimised guide RNA library that allows a 3-fold reduction in size and cost of genome-wide CRISPR-Cas9 screen, within the framework of a larger project aimed to understand to which extent gene essentiality varies in healthy humans. This new minimised libraries required the development of a new protocol for the sequencing of the amplicon from genomic DNA. The aim of this pilot is to verify that the new protocol performs well and is scalable.
