Comparison of the transcriptional profiles of male E13.5 wild type and L/H-Pgds-/- gonads

The goals of this study are to compare transcriptome profiling of L/H-Pgds-/- gonads to that of WT (RNA-seq) Methods: mRNA profiles of E13.5 wild-type (WT) and L- and H- prostaglandin D synthases (L/H-Pgds-/-) embryonic gonad were generated by deep sequencing, in triplicate, using Illumina HiSeq2000. qRT-PCR validation was performed using TaqMan and SYBR Green assays本研究旨在通过RNA测序（RNA-seq）技术，对比L型与H型前列腺素D合酶敲除（L/H-Pgds-/-）性腺与野生型（WT）性腺的转录组谱。实验方法如下：针对胚胎发育第13.5天（E13.5）的野生型（WT）及L、H型前列腺素D合酶敲除（L/H-Pgds-/-）胚胎性腺的mRNA，采用Illumina HiSeq2000测序平台进行深度测序，且设置三次生物学重复。后续通过TaqMan探针法与SYBR Green染料法开展实时定量反转录聚合酶链反应（qRT-PCR）验证实验。
