The Burmese amber specimen studied here originates from amber mines near the Noije Bum Hill (26°20' N, 96°36' E), Hukawng Valley, Kachin State, northern Myanmar. The specimen is deposited in the Nanjing Institute of Geology and Palaeontology (NIGP), Chinese Academy of Sciences, Nanjing, China. The amber piece was trimmed with a small table saw, ground with emery papers of different grain sizes, and finally polished with polishing powder. Photographs under incident light were taken with a Zeiss Discovery V20 stereo microscope. Widefield fluorescence images were captured with a Zeiss Axio Imager 2 light microscope combined with a fluorescence imaging system. Confocal images were obtained with a Zeiss LSM710 confocal laser scanning microscope. Images under incident light and widefield fluorescence were stacked in Helicon Focus 7.0.2 or Zerene Stacker 1.04. Confocal images were manually stacked in Adobe Photoshop CC. Images were further processed in Adobe Photoshop CC to enhance contrast.
本研究所用的缅甸琥珀标本采自缅甸北部克钦邦胡康河谷Noije Bum山(北纬26°20′,东经96°36′)附近的琥珀矿场,该标本馆藏于中国南京的中国科学院南京地质古生物研究所(Nanjing Institute of Geology and Palaeontology, NIGP)。该琥珀块先经小型台锯裁切修整,再以不同粒度的砂纸打磨,最终使用抛光粉完成抛光处理。入射光照明下的显微照片通过蔡司Discovery V20体视显微镜拍摄;宽场荧光图像由搭载荧光成像系统的蔡司Axio Imager 2光学显微镜采集;共聚焦图像则通过蔡司LSM710共聚焦激光扫描显微镜获取。入射光及宽场荧光图像均使用Helicon Focus 7.0.2或Zerene Stacker 1.04软件进行景深堆叠;共聚焦图像采用Adobe Photoshop CC软件手动完成堆叠。所有图像后续均通过Adobe Photoshop CC进行对比度增强处理。
