Transcriptome profiling of intestinal organoids stimulated with TNF and PGE2

To address the effect of inflammatory stimulation on intestinal epithelial cells, we performed RNA-seq analysis on human colonic epithelial organoids treated with TNF and/or PGE2. Intestinal organoids were cultured for 4 days and treated with TNF (10 ng/ml) with or without PGE2 (1 µM), or treated with PGE2 alone for 48 h. RNA was isolated (n = 3/group), and RNA-seq analysis was performed for the four different groups (Control/TNF/TNF+PGE2/PGE2).
