Using a murine model where early life skin colonization by Staphylococcus epidermidis leads to establishment of antigen-specific immune tolerance and S. epidermidis-specific regulatory T cells, we asked whether this response is perturbed by subsequent gut inflammation. Initiation of DSS-induced colitis in tandem with skin tape-stripping and S. epidermidis re-colonization in adult mice exposed to S. epidermidis at birth led to reduced commensal-specific Tregs in skin, skin-draining lymph nodes and gut-draining lymph nodes. To better understand the impact of colitis on composition of the colonic and skin bacterial communities in this model with performed 16S rRNA sequencing of skin swab and stool samples from mice in two separate experiments.
