We developed a chemical precision tool that allows cell-type specific labelling of glycoproteins. Cells are endowed with an artificial biosynthetic pathway that allows the synthesis of modified UDP-GalNAc when cells are fed with modified GalNAc precursor (Ac4GalN6yne). The aim of this RNA-seq analysis is to establish if the transfection of cells with the artificial biosynthetic pathway (NahK and AGX1-F383A enzymes) and/or the feeding treatment (DMSO or Ac4GalN6yne or Ac4GalNAc) have a substantial effect on the RNA transcriptomics. Comparative gene expression profiling analysis of RNA-seq data for k562 cells transfected with either empty or Nahk/AGX1_F383A plasmid at different feeding regimes.
