Synthetic tunable promoters for flexible control of multi-gene expression in mammalian cells

We reported the synthesis and expression measurement of ~50,000 tunable synthetic promoters. Each synthetic promoter contains a unique 12bp specific sequence at its terminal end, serving as a barcode that was uniquely transcribed into corresponding RNA in cells. After transfection of the pooled promoter library into 293T cells, RNA and DNA were extracted from the transfected cells for RNA-seq and DNA-seq, respectively. The normalized strength of each synthetic promoter was determined by dividing the RNA levels of each barcode by its corresponding DNA-seq levels.
