Proteomic data from nephron progenitor cells (untreated, YN1-treated, CHIR-treated)

Protein extracts from isolated NPC (treated and untreated) were reduced, alkylated and digested overnight. Samples are labeled with the TMT Reagents and then mixed before sample fractionation and clean up. Labeled samples are analyzed by high-resolution OrbitrapLC-MS/MS before data analysis to identify peptides and quantify reporter ion relative abundance.

从经处理与未处理的分离核孔复合物（Nuclear Pore Complex, NPC）中提取的蛋白质提取物，先经还原、烷基化处理并于过夜条件下酶解。样品采用TMT试剂（Tandem Mass Tag）标记后混合，随后进行样品分级分离与净化处理。标记后的样品通过高分辨率Orbitrap液相色谱-串联质谱（Orbitrap LC-MS/MS）分析，后续开展数据分析以鉴定肽段并定量报告离子的相对丰度。
