This study leverages ATAC-seq to reveal Arid3a's chromatin accessibility effects in VSMCs, identifying key gene expression changes and implicating Arid3a in actin filament dynamics linked to AAA pathogenesis. Overall design: We utilized RNA interference technology to knockdown Arid3a in Vascular Smooth Muscle Cells (VSMCs), successfully establishing several Arid3a knockdown cell lines. In this study, we applied ATAC-seq to dissect the chromatin accessibility dynamics in Vascular Smooth Muscle Cells (VSMCs) following Arid3a knockdown. The ATAC-seq assay was designed to capture the open chromatin regions that are potentially bound and regulated by Arid3a, offering insights into its direct and indirect transcriptional regulatory effects.
