The multiple intestinal neoplasia (MinApc delta716/+) enterotoxigenic Bacteroides fragilis (ETBF) murine colon tumorigenesis model is characterized by IL-17-dependent, distal colon adenomas. However, the addition of the human BRAFV600E to create BRAFV600ELgr5CreMin (BLM) to the ETBF model results in the addition of tumors in the mid-proximal colon as well as the infiltration of CD8 cells and INFg-producing cells, as observed by FACS analysis and IHC, in ETBF-induced BLM tumors compared to ETBF-induced Min tumors. We used RNA sequencing on BLM and Min tissues in order to better understand the changes in the colonic microenvironment induced by the addition of the BRAFV600E mutation.Overall design: We performed bulk RNA sequencing on mRNA isolated from sham tumors, sham epithelium, ETBF-induced tumors, and ETBF surrounding normal epithelium from both BLM and Min mice. We note that there is an increase in the expression of INFg pathways in BLM epithelium and ETBF-induced tumors of mice expressing the BRAFV600E mutation compared to those of Min mice.
