AAV2病毒变体小鼠肝脏组织感染能力文库Lib1,是包含约9.77万种AAV2变体的衣壳序列及其小鼠肝脏靶向分数的病毒文库。文库的设计方式是,在野生型AAV2衣壳蛋白561-588区域随机突变或在该区域内插入一个或多个氨基酸的随机多肽片段。依次进行(1)序列合成,(2)质粒构建,(3)在体外细胞培养皿中进行质粒转染以便包装病毒文库,(4)病毒文库测序,(5)病毒文库对小鼠给药,(6)小鼠肝脏组织取材和测序,(7)用自研的方法分析测序数据,从而得到该文库。最终该文库包含的数据条目有:AAV2病毒变体的碱基序列、氨基酸序列、病毒库中的病毒读数以及RPM、肝脏组织读数以及RPM、感染能力分数等。
The AAV2 variant library Lib1, designed to evaluate the infectivity of AAV2 variants in mouse liver tissue, is a viral library containing approximately 97,700 AAV2 variant capsid sequences and their corresponding mouse liver targeting scores. This library was constructed by either introducing random mutations into the 561-588 region of the wild-type AAV2 capsid protein, or inserting random polypeptide fragments consisting of one or more amino acids within this region. It was generated via the following sequential workflow: (1) sequence synthesis, (2) plasmid construction, (3) plasmid transfection in in vitro cell culture dishes to package the viral library, (4) viral library sequencing, (5) administration of the viral library to mice, (6) harvest of mouse liver tissue and subsequent sequencing, and (7) analysis of the sequencing data using a self-developed method. The final data entries included in this library cover: the nucleotide and amino acid sequences of the AAV2 variants, viral reads and RPM (Reads Per Million) from the viral stock, liver tissue reads and RPM, as well as infectivity scores and other related metrics.
