Gene expression analysis in peripheral blood mononuclear cells from newly diagnosed gestational diabetes mellitus

Gestational diabetes mellitus (GDM) is a metabolic disorder with adverse impact on maternal and fetal health. GDM generally starts form 24 weeks of gestation and persist till parturition (term). After termination of pregnancy, sugar comes to normal level and GDM aborts. However, the molecular mechanism involved in the progression and abortion of diabetic condition is not well known. The goal of this study was to compare the gene expression profiles of GDM mother and non-diabetic mother in order to elucidate the molecular mechanisms involved in it. Peripheral blood from Two GDM mother (before term), one GDM mother (after term) and one non-diabetic healthy control was withdrawn and total mRNA was extracted. Genome wide gene expression was compared with the help of MinION device and the data were validated by real time PCR. Differential expression analysis identified 859 differentially expressed genes (DEGs) and ---- differentially expressed transcripts. Of the 859 DEGs, 311 were up-regulated while 548 were down-regulated. GO and KEGG pathways enrichment analysis showed that these are associated with the insulin signaling pathway.
