This study proposes a paper/PMMA hybrid device designed to isolate exosomes and extract exosomal miRNA, followed by quantitative analysis. The paper/PMMA hybrid device with a microvalve-controlled design can provide simplified and convenient sample preparation for point-of-care testing (POCT) applications. The mechanical microvalve can exert high pressure on the paper, squeezing the porosity in the paper and thereby adjusting the flow rate and programming the fluid flow direction to make the extraction of miRNA successful. Here, we provide four Excel files that are the source files for plotting Figures 4-6 in the paper. The experimental data were recorded in these files. The file named "Flow Rate Test" shows the time of fluid flowed over the 5 distances on the paper pressed by the valve with different turns (0, 2, 5, and 7 turns) of the screw. The second file, PELISA, contains the records about the quantification analysis of paper-based ELISA. The third file, “Different samples for extraction", presents the Ct values for samples of two different volumes of cell culture media (CCM) solution and 10 uL of standard exosome solutions prepared in two concentrations. The fourth file, Extraction kit and the device, shows the Ct values for comparing the commercial extraction kit and the paper/PMMA hybrid device.
