Comparision of primay murine airway basal cells and ESC-derived basal cells

The goal of this experiment is to study the transcriptomic similarity and differences between ESC-derived cultured basal cells and their primary counterparts in the same culture condition. Overall design: We analyzed mouse mouse iBC culture from the directed differentiation protocol outlined in Ma et al., 2023. iBC cultures are previously sorted on Nkx2-1mCherry+/NGFR+ gate and cultured for 9 days in Basal Cell Medium. Meanwhile, we harvested airway basal cells from syngeneic adult mouse trachea and cultured them in the same BCM media. Both samples are outgrowth from NGFR+ FACS purified samples.
