Transcriptional profiles of liver lobular zones from non-pregnant mice and pregnant mice at dpc8 and dpc16.

We report transcriptional profiles of each liver lobular zone from non-pregnant mice and pregnant mice at dpc8 and dpc16. By RNA-sequencing of LCM-isolated each zonal cells, we revealed that genes related to actin cytoskeleton, extracellular matrix organization and vascularization were enriched in the PA (zone2) and CV (zone3) zones at dpc8 and cell division related genes were enriched in the PV (zone1) zone at dpc8. At dpc16, genes related to metabolism, catabolism and xenobiotic process were enriched in all zones. LMD-RNAseq analyses of liver lobular zones (zone 1,2, and 3) from non-pregnant mice and pregnant mice at dpc8 and dpc16.

本研究报道了未妊娠小鼠以及妊娠第8天(dpc8)、妊娠第16天(dpc16)小鼠肝脏各肝小叶带的转录组谱。通过对激光捕获显微切割(Laser Capture Microdissection, LCM)分离得到的各带细胞进行RNA测序(RNA-sequencing),本研究发现:妊娠第8天时,肌动蛋白细胞骨架、细胞外基质组织及血管生成相关基因在PA(zone2)与CV(zone3)区域富集,细胞分裂相关基因则在PV(zone1)区域富集。妊娠第16天时,代谢、分解代谢及异生物质代谢过程相关基因在所有肝小叶带中均出现富集。本研究同时对未妊娠小鼠以及妊娠第8天、妊娠第16天小鼠的肝小叶带(zone1、zone2、zone3)开展了LMD-RNAseq分析。
