the goal of this project is to demonstrate the functional heterologous complementation of DNA repair enzymes between two Thermococcales species. The model species are Thermococcus barophilus and Pyrococcus abyssi. Two strains of T. barophilus were genetically modified, respectively UBOCC-M-3302 (deletion of TERMP_RS08040 encoding PolB) and UBOCC-M-3303 (deletion of the locus TERMP_RS03345) and complemented by their homologous genes present on P. abyssi genome (PAB_RS09320 and PAB_RS02765, respectively encoding PolB and Rnase HII). The complementations were undertaken at a specific locus of by homologous recombination)
