Mesenchymal stem/stromal cells (MSCs) demonstrate great therapeutic potential in regenerative medicine. However, the differences between the transcriptome of MSCs from umbilical cord tissue and in vitro cell culture at single-cell level remain elusive. We obtained a large-scale single-cell transcriptome of total 19221 single cells from human umbilical cord Wharton's jelly tissue and the matched culture-expanded MSCs from three donors to provide a high quality transcriptomic profiling dataset at single-cell resolution, which is a valuable resource for dissecting the MSCs heterogeneity, cell fate decision in native tissue niche and in vitro culture environment for specific lineages.

间充质干/基质细胞(Mesenchymal stem/stromal cells, MSCs)在再生医学领域展现出优异的治疗潜力。然而,在单细胞分辨率下,脐带组织来源的MSCs与体外培养的同类型细胞的转录组差异仍未得到清晰阐释。本研究从3名供者的人脐带华通氏胶(Wharton's Jelly)组织及匹配的体外扩增MSCs中,共计获取了19221个单细胞的大规模转录组数据,构建了高质量的单细胞分辨率转录组图谱数据集。该数据集可为解析MSCs的细胞异质性、原生组织微环境及体外培养环境下的特定细胞谱系的细胞命运决定机制提供极具价值的研究资源。
