In order to identify the mechanism of action of cytotoxic dialkynylcarbinols (DACs), human pseudo-haploid HAP-1 cells were mutagenized with EMS and the mutagenized population was selected with a lethal 0.25 uM concentration of a (S)-DAC. Individual clones were isolated (DACR clones) and four of them, plus the wild-type HAP-1, were submitted to RNA-seq to identify mutations conferring resistance to (S)-DAC.
