Temperature sensitive (TS) mutants in the budding yeast strain S288C were crossed to 10 wild yeast isolates. Haploid segregant progeny carrying the TS allele were selected at both permissive and restrictive temperatures and segregant pools were sequenced to identify regions in the wild genome that carried a variant that could suppress the temperature sensitivity.
